His television debut came in 2015 when he joined the cast of the SABC3 soapie Isidingo, portraying Sibiya, a loyal, humorous, and outspoken miner. His portrayal of this character until 2020 earned him widespread popularity.

In 2017, Mtsweni appeared in the TV series iNumber Number as Nyoka, a cunning and ruthless gangster. He also portrayed an abducted boy on Rhythm City, another soapie on e.tv3. In 2020, he joined the cast of the telenovela Isibaya as Mpihlangene Zungu, the younger brother of the main protagonist, Mpiyakhe Zungu. His acting repertoire expanded in 2021 when he joined Gomora as Bongani.

Beyond television, Thulani Mtsweni has showcased his acting prowess in the short film Shaka Inkosi YamaKhosi, released in 2022. He played the role of Sompisi on the hit series Shaka Ilembe. His exceptional performance in the acting industry was recognized in 2023 when he won a South African Film and Television Award (SAFTA) in the Best Supporting Actor in a Telenovela category.
